The Effect of Human Thymus Fat-derived Mesenchymal Stem Cell on Myocardial Infarcted Rat Model

Purpose : Many patients develop heart failure following a myocardial ischemic event. In this light, stem cell therapy has shown promising results in cardiovascular disease. Thymic fat tissue is very familiar to cardiac surgeons due to routine removal of thymic tissue when undergoing sternotomy, but its uses in terms of stem cell therapy has not been well studied before. We assessed the effectiveness of mesenchymal stem cell (MSC) derived from thymic fat tissue in rats.
Methods : Human thymic fat tissues were obtained from the patients with ischemic heart disease who were undertaken coronary artery bypass graft surgery.
*Cell study : cumulative population doubling level,FACS analysis,and differentiation study.
*Animal study : We used SD rat(ischemia-reperfusion model) divided into three groups-treatment group(12 rats),vehicle group(11 rats),sham group(10 rats)
Trans-thoracic echocardiogram was used to assess the recovery of heart function including diastolic and systolic function every 1,4 weeks after surgical manipulations in each group.
Results : Compared with vehicle and treatment groups in terms of systolic and diastolic function of heart, systolic function between two groups were not significantly different at any point.However, diastolic function between vehicle and treatment groups showed relatively different results.After one week of stem cell or vehicle injection, treatment group showed statistically significant improvement in E/E(25.75±1.09 vs 24.20±0.91,P<0.001).But this trend was not consistent in four weeks.
Conclusion : It is impetuous to conclude the effect of MSC derived from thymic fat tissue in MI, but there is a trend in improvement in diastolic function of heart.Further research is needed to establish the effectiveness of thymic fat MSC.
